They do show ads in the form of Tweets from time to time. Easily blocked though.

They sell access to the firehose (the full stream of tweets from all users across the site or a % there of). A previous startup I worked in considered paying for a piece of it to track trends and sentiment in a real time manner.

What’s fun is that because of the recently passed Jobs Act they are required to release much less in the way of due diligence than they otherwise would have. In other words, now they don’t have to tell you where they money comes from before offering themselves up for investment.
